Simultaneous targeting of two different molecules in cancer is an effective way to shrink tumors, block invasion, and stop metastasis, scientists at the University of California, San Francisco (UCSF) have found?work that may improve the effectiveness of combination treatments that include drugs like Avastin.

The two-target approach, tested in mice with a type of cancer known as neuroendocrine pancreatic tumors, may have broad application for treating a wide variety of cancers, the UCSF team said. The drugs used in the tests belong to classes of pharmaceuticals that are either on the market or under development in clinical trials.

Clinical trials also are already underway to gauge effectiveness of the approach in humans with prostate cancer, breast cancer, and other tumor types. The UCSF study, described in the journal Cancer Discovery this week, is the first to show how the drug combination works in the laboratory.

The results are promising, said Donald McDonald, MD, PhD, a member of the UCSF Helen Diller Comprehensive Cancer Center and the Cardiovascular Research Institute and professor of anatomy, who led the research.

In the study, treating mice with the dual-target approach turned aggressive tumors with invasive fingers penetrating surrounding tissues and many metastases into tiny balls with few or no metastases.

"It's the combination of approaches?there's a synergy between the two," McDonald said. "You add two and two, and you get 10."

How each target works

The two targets are both proteins that scientists have known for years are involved in cancer. Both play important roles in malignant tumors.

The first, called c-MET, is involved in two processes associated with the most deadly cancers. A clinical marker of cancer aggressiveness, c-MET drives tumor invasion into surrounding tissues. It is also involved in metastasis?the spread of cancer cells to other parts of the body where they can establish new tumors.

The second target is a protein known as vascular endothelial cell growth factor (VEGF). VEGF is a protein that promotes the growth of new blood vessels. Growing tumors hijack this process to expand their network of blood vessels to provide nutrients. Drugs blocking VEGF have been developed based on the simple assumption that tumors cannot grow if you choke off their blood supply.

Drugs that target these molecules are in development, and a few are already on the market. The U.S. Food and Drug Administration (FDA) approved the first of these in 2004 to treat metastatic colon cancer. That drug, called Avastin, is manufactured by the South San Francisco-based company Genentech. Avastin was approved for metastatic breast cancer in 2008 under the FDA's accelerated approval program.

The FDA revoked approval of Avastin for breast cancer last year after further assessing the relative risks and benefits to women taking it. Blocking VEGF seemed to slow tumor growth for awhile, but the FDA determined that it did not significantly improve or extend the lives of most women taking it.

"It was not clear why some tumors responded and others did not. It was also unclear why some tumors would respond initially and then would stop responding," said McDonald, who has studied blood vessels in tumors and the effect of cancer drugs for years in his UCSF laboratory.

Two years ago former UCSF professor Douglas Hanahan and colleagues found in laboratory experiments that Avastin-like drugs would shrink tumors but unexpectedly did something else as well. The drugs also morphed tumors from roundish blobs into highly irregular growths with tendrils that penetrated surrounding tissues and even spread to other organs?suggesting that the VEGF blockade could also make tumors more aggressive, invasive and metastatic.

McDonald's group confirmed Hanahan's findings and discovered that c-MET was involved. In their latest research, Barbara Sennino, PhD, with other investigators in his group set out to determine whether c-MET drove tumor aggressiveness during anti-VEGF therapy. What their paper shows is that blocking c-MET and VEGF together in mice is more powerful than blocking either alone because it not only slows tumor growth but also reduces invasion and metastasis.

They tested two inhibitors of VEGF?a neutralizing antibody and sunitinib?and three inhibitors of c-MET?crizotinib, PF-04217903, and cabozantinib (XL184). Unlike the other agents, cabozantinib simultaneously inhibits both c-MET and VEGF. Inhibition of c-MET and VEGF together with a drug combination or with cabozantinib had more profound effects on tumors than any of the agents that blocked only one of the targets.

These promising laboratory results still need more tests of safety and effectiveness in the clinic, McDonald said, and it may be a year or more before the drugs are routinely available to patients.

With a name like that, it's not surprising that months of research by the experts at New Mexico State University's Chile Pepper Institute have identified the variety as the new hottest pepper on the planet.

The golf ball-sized pepper scored the highest among a handful of chile breeds reputed to be among the hottest in the world. Its mean heat topped more than 1.2 million units on the Scoville heat scale, while fruits from some individual plants reached 2 million heat units.

"You take a bite. It doesn't seem so bad, and then it builds and it builds and it builds. So it is quite nasty," Paul Bosland, a renowned pepper expert and director of the chile institute, said of the pepper's heat.

Researchers were pushed by hot sauce makers, seed producers and others in the spicy foods industry to establish the average heat levels for super-hot varieties in an effort to quash unscientific claims of which peppers are actually the hottest.

That's something that hadn't been done before, Bosland said.

"The question was, could the Chile Pepper Institute establish the benchmark for chile heat?" he said. "Chile heat is a complex thing, and the industry doesn't like to base it on just a single fruit that's a record holder. It's too variable." The academic institute is based at the university's agriculture school and is partially funded by federal grants, as well as some industry groups depending on the project.

The team planted about 125 plants of each variety ? the Trinidad Moruga Scorpion, the Trinidad Scorpion, the 7-pot, the Chocolate 7-pot and the Bhut Jolokia, which was a previous record-holder identified by the institute and certified by Guinness World Records in 2007.

Randomly selected mature fruits from several plants within each variety were harvested, dried and ground to powder. The compounds that produce heat sensation ? the capsaicinoids ? were then extracted and examined.

During harvesting, senior research specialist Danise Coon said she and the two students who were picking the peppers went through about four pairs of latex gloves.

"The capsaicin kept penetrating the latex and soaking into the skin on our hands. That has never happened to me before," she said.

Chile peppers of the same variety can vary in heat depending on environmental conditions. More stress on a plant ? hotter temperatures or less water, for example ? will result in hotter fruit.

The Trinidad Moruga Scorpion's new notoriety is already making waves in the industry and among those who love their hot, spicy foods.

"As with all the previous record holders, there will be a run on seeds and plants," said Jim Duffy, a grower in San Diego who supplied the university with seeds for four of the super-hot varieties. "Like Cabbage Patch dolls right before Christmas or Beanie Babies, it's like the hot item."

Not even Duffy or the researchers would dare to pop a whole Trinidad Moruga Scorpion in their mouths, but there are plenty of videos on social networking sites where heat-loving daredevils have tried.

The blood flow increases and the endorphins start flowing. Their faces turn red, the sweat starts rolling, their eyes and noses water and there's a fiery sensation that spreads across their tongues and down their throats.

"People actually get a crack-like rush," Duffy said. "I know the people who will eat the hottest stuff to get this rush, but they've got to go through the pain."

Pepper experts said there are a handful of people who are crazy enough to subject themselves to the pain, but the rest just want to try out these super-hot peppers on their friends or make killer hot sauce ? and it doesn't take a whole pepper to do that.

More bang for the buck is how Bosland describes it. He said a family could buy two of the super-hot peppers to flavor their meals for an entire week.

The beauty of the peppers is they're not only the hottest in the world, but they're also some of the most flavorful peppers, Duffy said.

"You can make a barbeque sauce or a hot sauce at a mild to medium level using small amounts of these peppers and it will be so darn addictive that you won't want to put your spoon down," he said. "You'll want to eat and eat and eat."

TORONTO (Reuters) ? In another blow for beleaguered BlackBerry maker Research In Motion, the U.S. federal government's main procurement agency is issuing iPhones and Android-based devices to some of its 17,000 workers.

Oilfield services company Halliburton plans to switch 4,500 BlackBerry-toting employees to iPhones, saying it's is better suited to its needs. Several banks have also welcomed BlackBerry rivals.

The NOAA move was made possible after it switched its desktop-based software to Google Apps for Government.

Coleman said the GSA moved to Google Apps in June, cutting its costs in half compared with its legacy desktop software.

Novice: The Gananoque Novice Islanders are headed in the right direction and rebounded on weekend with two wins.

The wins give the Islanders a 4-2 record in post season action with six games remaining to play in playdowns.

Gananoque took on Loyalist last Thursday and squeaked out a 2-1 victory. Wil Murphy and Wyatt Bailey scored the two Islanders goals for the win.

Then on Sunday, the team travelled to Oshawa and trounced the home team 8-1.

?There was more determination from the kids,? said coach Lanny Murphy of the weekend's performance. ?We had steady defence and we need to keep that strong effort in the last six games if we're going to finish in the top two spots.?

Bailey and Murphy both had three goals in the victory, while Jordan Gillis and Clayton Rose both had lone goals.

?We're headed in the right direction,? said Murphy.

It's another two game weekend for Gananoque as they play in Ennismore on Saturday before returning home on Sunday to take on Oshawa again at 3:15 p.m. at the Lou Jeffries Gananoque TLTI Recreation Centre.

Peewee: The Gananoque Peewee Islanders came back to tie their second round series at one game apiece after defeating Ennismore 3-1 on Sunday.

With all Islander skaters back in action for game two, the match was set to be a battle. Ennismore, with home ice advantage, came out strong and was a dominant presence on the ice for the first period.

Donovan ripped a shot that left a rebound and a scramble ensued in front of the net. Matthew Micallef banged at the loose puck and it deflected off Kinsella's stick and in for a 2-1 lead.

Less than forty seconds later, Ben Cartwright sent the puck back to the point for Logan Leonard. Leonard ripped his own shot that Hayden deflected five-hole and the Islanders had a 3-1 advantage.

The fifth game is back in Ennismore on Sunday, with a possible sixth game scheduled for 7 p.m. Tuesday in Gananoque.

Bantam: The Gananoque Bantam Islanders were eliminated from the playoffs in three straight games at the hands of Ennismore.

The Islanders first were handed a 3-0 loss in Ennismore on Saturday to put them down two games to none in second round playoff action.

THURSDAY, Feb. 9 (HealthDay News) -- Heart disease may be a risk factor for prostate cancer, a new study suggests.

If this link is confirmed in future research, it means that lifestyle changes that reduce heart disease risk -- such as weight loss, exercise and a healthy diet -- may also protect men against prostate cancer, the Duke Cancer Institute researchers said.

"What's good for the heart may be good for the prostate," study author Dr. Jean-Alfred Thomas II, a postdoctoral fellow in the division of urology, said in a Duke Medicine news release.

He and his colleagues analyzed data from 6,390 men in a four-year clinical trial testing a drug's effectiveness in reducing prostate cancer risk. Of those men, 547 reported a history of coronary artery disease before the start of the trial.

The Duke researchers found that men with coronary artery disease had a 35 percent greater risk of developing prostate cancer over time and a 24 percent greater risk of being diagnosed with prostate cancer within the first two years of the study compared to men who did not have heart disease.

Four years into the clinical trial, men with coronary artery disease had a 74 percent higher risk of prostate cancer than those with no heart disease.

"We controlled for a number of risk factors, including hypertension, taking statins or aspirin," Thomas said. "We don't have a good grasp on what's causing the link, but we are observing this association."

The study appears online this month in the journal Cancer Epidemiology, Biomarkers & Prevention.

Coronary artery disease is the leading cause of death among adults in the United States, and prostate cancer is the second most deadly type of cancer for men in the United States, the release noted.

TUESDAY, Feb. 7 (HealthDay News) -- The risk of dying from a hormone receptor-positive breast cancer increases with age, according to new research. And one reason might be that older women with breast cancer are undertreated compared to their younger peers.

For women between the ages of 65 and 74, the risk of dying from breast cancer was 25 percent higher than for women under 65. For those 75 and older, the risk of death was 63 percent higher than for women under 65, according to the new study.

"This study showed that among postmenopausal, non-metastatic, breast cancer patients, elderly patients have a higher risk of dying from breast cancer than younger patients," said study author Dr. Cornelis van de Velde, a professor of surgery at Leiden University Medical Center in the Netherlands and president of the European Cancer Organization.

Results of the study are published in the Feb. 8 issue of the Journal of the American Medical Association.

Breast cancer affects about 226,000 American women every year, according to the American Cancer Society. Worldwide, there were nearly 1.4 million new cases in 2008, according to background information in the study.

Breast cancer tumors can be either hormone receptor-negative or positive. If a tumor is hormone receptor-positive, that means that the tumor's growth is fueled by hormones, such as estrogen, progesterone or both. Certain treatments specifically target hormone receptor-positive cancers.

In the current study, researchers reviewed data from a previous trial of treatments designed for hormone receptor-positive breast cancers. The nearly 10,000 women included were randomly assigned to receive one of two drugs, exemestane (Aromasin) or tamoxifen (Nolvadex), for 2.5 to 3 years, and then all received Aromasin for an additional 2 to 2.5 years. The women were from nine different countries, and all had hormone receptor-positive breast cancer.

There were more than 5,300 women under age 65 in the study; over 3,000 women between the ages of 65 and 74; and just over 1,350 women above the age of 75.

Not unexpectedly, the overall risk of death from any cause was higher for older women. As women aged, the proportion of all-cause deaths attributed to breast cancer became smaller.

However, when the researchers looked at breast cancer-specific deaths, they found that older women were significantly more likely to die from breast cancer than younger women. The investigators also found that for every 10-year increase in age, the risk of death from breast cancer went up by 20 percent.

Van de Velde said the current study can't answer the reason why older women have a higher risk of death from breast cancer, but that "the most plausible explanation is that elderly patients may experience undertreatment."

Len Lichtenfeld, deputy chief medical officer at the American Cancer Society, commented on the findings.

"There has been an assumption that older women may have a more indolent course of breast cancer, and that there was more of a risk of overtreating older women," Lichtenfeld said. "This study suggests that the course of the disease isn't so indolent, and that breast cancer still plays a significant and increasing role in the risk of death in older women."

Lichtenfeld said that while the reasons aren't clear from this study, undertreatment is a possibility. He said there may be a concern about how well older women might handle the side effects of cancer treatments. It's also possible that there's something different in the breast cancer itself that may make it more aggressive, or breast cancers in older women may not respond as well to the available treatments.

Lichtenfeld also noted that other medications older women take could potentially interact with cancer treatments.

The study found that older women tended to have larger tumors at the time of diagnosis, which also could have affected their outcome. But, it wasn't clear if older women had larger tumors when diagnosed because of a lack of screening, or again, if it had something to do with the type of breast cancer they had.

ASLA Communications Internship, Summer 2012 ? The Dirt

The American Society of Landscape Architects (ASLA) seeks a full-time summer intern for an exciting project: The Landscape Architect?s Guide to Washington, D.C. This Web site, which will include both mobile-friendly version and a more robust online exhibition, will feature both well-known and up-and-coming landscape architects discussing what makes 50 landscapes within the nation?s capital compelling. The Web site is expected to launch in fall 2012.

Instead of offering yet another brochure for tourists, the site is designed to be a guide to both landscapes and design-thinking. The goal of the project is to educate the?milion of visitors who come to?D.C.?about how landscape architects, designers of public spaces, critically evaluate the design of sites. Landscape architects will discuss the site plans, design details, interesting historic features, and sustainable design elements. Landscapes featured will include great works of landscape architects (the National Mall, Dumbarton Oaks), ecological landscapes, historic landscapes, and even post-industrial urban landscapes. Local D.C. residents and landscape architects and other design professionals are also?target audiences for the site.

Responsibilities:

The summer intern will be expected to work full-time on this project from June through August.

The intern will research and write an introductory paragraph on the site?s history, using historical records and available books and Web sites; locate photographs, secure image credits, and add directional information for each site; coordinate outreach materials to ASLA members and aid in social media promotion; and directly interact with a number of leading landscape architects to gather their feedback on given sites and edit the text for publication.

Interns will also have the opportunity to attend educational and networking events at the National Building Museum, Harvard University?s Dumbarton Oaks, and other museums and think tanks in Washington, D.C. and write articles for ASLA publications including The Dirt blog and LAND newsletter.

Required Skills:

• Current enrollment in a Master?s or PhD program in landscape architecture.
• Excellent writing skills. The intern must be able to write clearly for a general audience.
• Proven research skills and ability to quickly evaluate the quality and relevance of many different types of Web resources.
• Excellent interpersonal skills and ability to interact graciously with busy designers.
• Working knowledge of Photoshop, social media platforms (Facebook, LinkedIn), and Microsoft Office suite.

Some of Canada's biggest acts will face off in the Album of The Year category: Avril Lavigne for Goodbye Lullabye, Drake for Take Care, Justin Bieber for his Christmas-themed Under the Mistletoe, Michael Bublé for his holiday album Christmas and rockers Nickelback for Here and Now.

Bieber scored his second nomination in the Fan Choice Award category along with Arcade Fire, Lavigne, City and Colour, deadmau5, Drake, Ginette Reno, Hedley, Bublé and Nickelback. He, however, didn't manage to land any nods in other major categories like Artist of the Year.

His pal Drake did nab a nom in the Artist of the Year category, along with Feist, Bublé, City and Colour and Deadmau5. While the roster of nominees for the show is uniquely Canadian, Drake's Stateside crossover makes him the most high-profile nom in the Rap Recording of the Year category. His Take Care will face off against Classified's Handshakes and Middle Fingers, D-Sisive's Jonestown 2: Jimmy Go Bye Bye, Kardinal Offishall's Anywhere (Ol Time Killin Part II) and Swollen Members' Dagger Mouth.

Nickelback, Feist and Deadmau5 are among the artists set to perform at the show, which takes place on April 1.
